Diabetes mellitus remains a major global health concern, prompting the exploration of traditional medicines as potential therapeutic agents. This study aims to evaluate and compare the in vitro antidiabetic activity of two Ayurvedic formulations: the commercially available Divyamadhunashini Vati and a newly developed in-house herbal blend. Both formulations were assessed for their ability to inhibit carbohydrate-digesting enzymes, specifically α-amylase and α-glucosidase, using established in vitro methods. Antioxidant activity was also measured through DPPH radical scavenging assays to examine their potential in reducing oxidative stress, a key factor in diabetes progression. The results revealed that both formulations possess notable enzyme inhibitory and antioxidant properties, with the in-house formulation showing slightly superior performance in α-glucosidase inhibition and free radical scavenging capacity. These findings suggest that the in-house preparation may serve as an effective natural alternative for managing hyperglycemia. Further in vivo and clinical evaluations are recommended to validate its therapeutic potential.
